| ASCT2（又稱SLC1A5）是溶質(zhì)載體家族1A中的一種鈉依賴性中性氨基酸反向轉(zhuǎn)運體。它的核心功能是以谷氨酰胺為首選底物，與細胞內(nèi)其他中性氨基酸（如絲氨酸、蘇氨酸）進行交換，以此調(diào)節(jié)氨基酸穩(wěn)態(tài)。 ASCT2之所以成為癌癥研究的熱門靶點，是因為它在多種腫瘤細胞中顯著高表達，為癌細胞快速增殖所必需的谷氨酰胺代謝提供原料。其結(jié)構(gòu)與功能機制復雜，最新研究發(fā)現(xiàn)其轉(zhuǎn)運循環(huán)中可結(jié)合三個鈉離子，且由于在關(guān)鍵跨膜結(jié)構(gòu)域上缺乏變構(gòu)能力，使其被限定在交換模式而非主動積累模式。 | |||
