受體酪氨酸激酶在正常細胞功能中的作用至關重要,人類有58 種已知的受體酪氨酸激酶,該家族最重要的成員之一是c-KIT。c-KIT 也稱為CD117,是一種由KIT 基因編碼的III 型受體酪氨酸激酶。它以干細胞生長因子(SCF)為配體,因此被稱為干細胞生長因子受 體(SCFR)。c-Kit 的信號轉(zhuǎn)導通過其配體SCF 蛋白的結(jié)合而被激活,通過自磷酸化引起 c-Kit 二聚化和激活,并啟動磷酸化級聯(lián)反應,從而調(diào)節(jié)細胞生長。c-Kit 磷酸化可觸發(fā)多種 信號轉(zhuǎn)導通路,包括JAK/STAT、RAS/MAP 激酶通路、PI3 激酶、PLCγ通路和SRC 通路。 SCF/c-KIT 信號轉(zhuǎn)導通路的激活通常與細胞增殖、遷移和存活有關,從而調(diào)節(jié)造血、色素沉著和精子發(fā)生的關鍵功能。 |

SCF/c-KIT(GNNK-)Effector Reporter Cell,在由調(diào)控因子調(diào)控并表達報告基因的重組細胞上,穩(wěn)定表達人c-KIT(GNNK-) 。SCF/c-KIT(GNNK-)Effector Reporter Cell 報告基因藥靶模型很好的模擬了體內(nèi)SCF/c KIT 的信號轉(zhuǎn)導過程,原理見下圖所示。
