| 腫瘤壞死因子-α(TNF-α)是一種分子量為26 kDa 的跨膜蛋白,其前肽在TNF-α-轉(zhuǎn)換酶(TACE)裂解時(shí)以可溶性17 kDa 分子的形式分泌。雖然活化的巨噬細(xì)胞是TNF-α的主 要來(lái)源,但它也可以由多種其他細(xì)胞產(chǎn)生,如成纖維細(xì)胞、星形膠質(zhì)細(xì)胞、平滑肌細(xì)胞、 角化細(xì)胞和多種腫瘤細(xì)胞,包括B 細(xì)胞淋巴瘤、乳腺癌和結(jié)腸癌。 TNF-α作為腫瘤促進(jìn)因子,參與腫瘤發(fā)生的所有步驟,包括腫瘤的轉(zhuǎn)化、增殖、血管生成、侵襲和轉(zhuǎn)移。TNF α信號(hào)傳導(dǎo)也是免疫系統(tǒng)的重要組成部分,可抑制腫瘤發(fā)生,阻止病毒復(fù)制,是一種內(nèi)源性熱原,可誘導(dǎo)發(fā)燒和細(xì)胞凋亡。 |
