| I. Background | |
| TPOR即血小板生成素受體（Thrombopoietin Receptor），由MPL原癌基因編碼，也稱為CD110、c-Mpl或MPL。它屬于I型細胞因子受體家族，是調(diào)控造血干細胞自我更新、巨核細胞分化和血小板生成的核心受體。其配體TPO結(jié)合后，TPOR發(fā)生同源二聚化并激活下游JAK2激酶，進而啟動STAT、PI3K等信號通路。該受體約635個氨基酸，分子量約71 kDa，廣泛表達于造血干細胞、巨核細胞及血小板表面。在臨床上，TPOR是重要的藥物靶點，相關(guān)激動劑（如艾曲泊帕、羅米司亭）已用于治療血小板減少癥，而其基因突變則與骨髓增殖性腫瘤及家族性血小板增多癥相關(guān)。 | |
