ADCP(Antibody-dependent cellular phagocytosis,抗體依賴性細胞介導的吞噬作用)是一種重要的免疫清除機制。治療性單克隆抗體(mAb)結合靶細胞后,其Fc段可被吞噬免疫細胞(如巨噬細胞、單核細胞、中性粒細胞)表面的Fcγ受體(FcγR)識別,從而介導對靶細胞的吞噬清除。 在人類中,F(xiàn)cγR家族包含激活型受體(如FcγRIIa/CD32a、FcγRI/CD64、FcγRIIIa/CD16a)和抑制型受體FcγRIIb(CD32b)。FcγRIIb是唯一表達于免疫細胞表面的抑制性Fcγ受體,其胞內段含有一個免疫受體酪氨酸抑制基序(ITIM)。當FcγRIIb與抗體Fc段結合并被交聯(lián)時,ITIM被磷酸化,招募含SH2結構域的肌醇磷酸酶(SHIP),進而抑制下游激活信號通路,從而負向調控ADCP及其他效應功能。FcγRIIb在維持免疫穩(wěn)態(tài)、防止過度炎癥反應中發(fā)揮關鍵作用,同時也是腫瘤免疫治療和抗體藥物研發(fā)中重要的潛在靶點。 |
