| I. Background | |
| APRIL（增殖誘導(dǎo)配體，官方基因名 TNFSF13，亦稱 CD256）是腫瘤壞死因子超家族 成員，由 17 號染色體 TNFSF13 基因編碼，屬Ⅱ型跨膜蛋白，經(jīng)蛋白酶解后以同源三聚體形 式分泌。APRIL 通過高親和力受體 BCMA 和低親和力受體 TACI 發(fā)揮作用，主要由髓系細(xì) 胞、活化淋巴細(xì)胞及黏膜上皮細(xì)胞產(chǎn)生，核心功能是促進(jìn) B 細(xì)胞向漿細(xì)胞分化并維持其存活，尤其在黏膜部位驅(qū)動 IgA 類別轉(zhuǎn)換，是長壽漿細(xì)胞的關(guān)鍵生存因子。 | |
| II. Description | |
APRIL CHO 細(xì)胞，在 CHO-K1 細(xì)胞的基礎(chǔ)上，穩(wěn)定表達(dá)人 APRIL。APRIL CHO 細(xì)胞作為 TACI Effector Reporter Cell 細(xì)胞的靶細(xì)胞，很好的模擬了體內(nèi)TACI&APRIL 的信號轉(zhuǎn)導(dǎo)過程，原理見下圖所示。 Figure 1. APRIL CHO 細(xì)胞模型原理圖 |
