| I. Background | |
| KLK2 基因（激肽釋放酶 2）位于人類 19 號染色體短臂（19q13.4），編碼一種膜結(jié)合的胰蛋白酶樣絲氨酸蛋白酶，分子量約 51 kDa。其結(jié)構(gòu)包含兩個六股β桶和五個二硫鍵， 底物結(jié)合口袋 S1 帶負電，可與肝素等調(diào)節(jié)因子結(jié)合，增強與蛋白酶抑制劑的相互作用。 KLK2 基因的表達受雄激素受體（AR）嚴(yán)格調(diào)控，啟動子區(qū)域含雄激素反應(yīng)元件（ARE）， 在雄激素刺激下，AR 二聚化并轉(zhuǎn)位入核，結(jié)合 ARE 啟動轉(zhuǎn)錄，因此 KLK2 特異性表達于前列腺上皮細胞及前列腺癌細胞，是前列腺癌的標(biāo)志物之一。 功能上，KLK2 能特異性切割 proPSA（前列腺特異性抗原前體）的 N 端肽鏈，激活其蛋白水解功能，參與精液液化及精子活動調(diào)節(jié)。同時，它通過激活 PSA 及促進血管生成， 增強前列腺癌細胞的侵襲性和轉(zhuǎn)移能力，其高表達與腫瘤侵襲性呈正相關(guān)。 |
