| I. Background | |
| TM4SF1 基因編碼的跨膜蛋白 4 超家族成員 1（TM4SF1）是一種四次跨膜糖蛋白，屬 于 Tetraspanin 超家族，在多種腫瘤中異常高表達(dá)。該蛋白通過四個疏水跨膜結(jié)構(gòu)域與整合 素、受體酪氨酸激酶等分子相互作用，參與細(xì)胞信號轉(zhuǎn)導(dǎo)、黏附和遷移，促進(jìn)腫瘤血管生 成、上皮-間充質(zhì)轉(zhuǎn)化（EMT）及腫瘤干細(xì)胞自我更新，顯著增強(qiáng)癌細(xì)胞侵襲和轉(zhuǎn)移能力。 臨床研究表明，TM4SF1 在肺癌、乳腺癌、肝癌、結(jié)直腸癌及卵巢癌等多種惡性腫瘤 中顯著上調(diào)，其表達(dá)水平與腫瘤轉(zhuǎn)移及不良預(yù)后密切相關(guān)。研究發(fā)現(xiàn)，TM4SF1 通過激活 JAK2-STAT3 信號通路，促進(jìn)乳腺癌向肺、骨、腦等遠(yuǎn)端器官轉(zhuǎn)移；同時，它還參與腫瘤 耐藥機(jī)制，降低治療效果。 |
